Simvastatin

Indications/Uses

CHD, hyperlipidemia. Adjunct to diet for reduction of elevated total & LDL cholesterol, triglyceride, & raises HDL.

Dosage/Direction for Use

CHD Initially 10-20 mg once every evening. Hyperlipidemia Initially 10-40 mg once every evening. Dose range: 5-80 mg daily. Concomitant bile acid sequestrant, cyclosporine, danazol Initially 5 mg once daily. Maintenance: Max of 10 mg daily. Concomitant gemfibrozil Max: 10 mg daily. Severe renal insufficiency Initially 5 mg once daily with close monitoring. Max: 10 mg daily.

Administration

May be taken with or without food: Avoid excessive consumption (>1 L/day) of grapefruit juice.

Contraindications

Active liver disease or unexplained persistent elevation of serum transaminases. Childn, pregnancy & lactation.

Special Precautions

Patient who consumes substantial amount of alcohol, history of liver disease.

Adverse Reactions

Abdominal pain, flatulence, constipation, asthenia & headache. Rarely, hepatitis, rhabdomyolysis.

Drug Interactions

Anticoagulants, cyclosporin, fibric acid derivative, itraconazole, ketoconazole, erythromycin, nefazodone, immunosuppressant.
